چکیده
Background: Cerebral ischemia is a common and serious neurological disease. Ephedra-cinnamomi composed of Ephedra and cinnamomi is a famous herb-pair in Traditional Chinese Medicine. The aim of present study was to investigate the effect of Ephedra-cinnamomi on cerebral ischemia injury in rats. Material and methods: Four-Vessel Occlusion (4-VO) method was applied to induce global cerebral ischemia injury. Rats were treated with Ephedra-cinnamomi (3, 6 and 12 g/kg, i.g.) while the untreated rates were regarded as the negative control. Then memory capacity was measured by Step through Passive Avoidance (STPA) test and Morris Water Maze (MWM) test. Pro-inflammatory cytokines levels were detected by Enzyme Linked Immunosorbent Assay (ELISA). Additionally, the protein expression levels of Toll-Like Receptor (TLR) 4, myeloid differentiation factor 88 (MyD88) and phosphorylated p38 mitogen-activated protein kinases (p-p38MAPKs) were detected by Western blot. Results: The results revealed that Ephedra-cinnamomi attenuated the cerebral ischemia-induced memory deficits and ameliorated histopathological changes. In addition, Ephedra-cinnamomi was able to reduce the pro-inflammatory cytokines production and inhibit the activation of TLR4/MyD88/p38 MAPK pathway in 4-VO induced rats model. Conclusion: The results indicated that Ephedra-cinnamomi significantly ameliorates the ischemic brain injury in rats. Furthermore, the molecular mechanism of improved ischemic brain by Ephedracinnamomi is associated with the inhibition of TLR4/p38 MAPK pathway and the suppression of proinflammatory cytokines production.

Contribution of p38 MAPK to the Ameliorating Effect of Enriched Environment on the Cognitive Deficits Induced by Chronic Cerebral Hypoperfusion.
BACKGROUND/AIMS An enriched environment (EE) ameliorates learning and memory impairments induced by chronic cerebral hypoperfusion, and the p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway exerts both beneficial and deleterious effects on the nervous system during the progression of ischemia. METHODS The present study investigated whether p38 MAPK participates in the process ...
متن کاملChronic intermittent hypobaric hypoxia attenuates monocrotaline-induced pulmonary arterial hypertension via modulating inflammation and suppressing NF-κB /p38 pathway
Objective(s): Inflammation is involved in various forms of pulmonary arterial hypertension (PAH). Although the pathophysiology of PAH remains uncertain, NF-κB and p38 mitogen-activated protein kinase (p38 MAPK) has been reportedto be associated with many inflammatory mediators of PAH. This study aimed to evaluate the effect of chronic intermittent hypobaric hypoxia (CIHH) on pulmonary inflammat...
متن کاملFructus mume alleviates chronic cerebral hypoperfusion-induced white matter and hippocampal damage via inhibition of inflammation and downregulation of TLR4 and p38 MAPK signaling
BACKGROUND Fructus mume (F. mume) has been used as a traditional medicine for many years in Asian countries. The present study was designed to determine the effect of a 70% ethanol extract of F. mume on white matter and hippocampal damage induced by chronic cerebral hypoperfusion. METHODS Permanent bilateral common carotid artery occlusion (BCCAo) was performed on male Wistar rats to induce c...
متن کاملExtracellular HSP60 induces inflammation through activating and up-regulating TLRs in cardiomyocytes.
AIMS The molecular events leading from cardiomyocyte ischaemia to inflammatory cytokine production are not well understood. We previously found that heat shock protein 60 (HSP60) appeared in extracellular space after cardiomyocyte ischaemia. This study examined the activation and regulation of toll-like receptors (TLRs) by HSP60 in cardiomyocytes. METHODS AND RESULTS Cytokine production and T...
متن کاملFerulic acid exerts neuroprotective effects against cerebral ischemia/reperfusion-induced injury via antioxidant and anti-apoptotic mechanisms in vitro and in vivo
Ferulic acid (FA) is a derivative of cinnamic acid. It is used in the treatment of heart head blood-vessel disease and exerts protective effects against hypoxia/ischemia-induced cell injury in the brain. This study investigated the potential neuroprotective effects of FA against ischemia/reperfusion (I/R)-induced brain injury in vivo and in vitro through hematoxylin and eosin (H&E) and Nissl st...
